Should the Major nutrient Stock for MS media be like water(transparent) or will it be Prepcipitated(Turbid white colour).
I found this precipitation only after i added calcium chloride..
kindly help me to solve this confusion..
Should be transparent as water. And yes, you are right..the culprit is CaCl2. In MS stocks the CaCl2 is always prepared as a 10-20 X stock separately and NOT mixed with MS major/ macro nutrients (which is 10 X sock for freshness, as long term storage would precipitate things too!). Same goes for Iron-EDTA as a separate 20-200 X Stock. Similarly, micronutrients and vitamins are different stocks too. But, you gauged it right. Certainly, NO CaCl2 to be mixed with MS Major stock. I totally agree with Biswapriya Biswavas Misra. Besides make sure that you used tissue-culture grade chemicals with high purity.
Alternatively all the chemicals of macro nutrients can be dissolved separately and then mixed. Iron-EDTA preparation should be as suggested by Biswapriya.
