Why using the whole-cell biosensor gives a fluctuative signal when detecting AHL ?

I've been working with E.coli JM109 pSB401 for detecting the C8-HSL from Burkholderia cenocepacia species. The AHL was extracted using Acidified Ethyl-acetate and then suspended into Methanol before used. The bioluminescenes did show up but the signal (RLU) wasn't stable for each cycle of reading (it's going up and down randomly). How can this happen ?? The signal was relatively low.

In "low" signal range, most spectroflurometers tend to have noise. If you are working in a range where measurements may be possible, try to ensure that there are absolutely no suspended insoluble material in your solution.
