Why there is no color conversion of purple color to yellow while working with bitter gourd fruits dry powder by DPPH assay??

It depends on the concentration of your samples, so try to increase the concentration to have significant antioxidant activity.
I am using 1g powder in 10ml MeOh and sonication for 60min and cfg for 10min@4000rpm then tried 100ul,100ul,400ul,500ul of sup and 3ml of 0.01mM of DPPH even no color .

Have you checked the pH?
In many cases the color change depends on pH change, so if you are using a buffer don't expect a color change. Usually, purple color is associated with higher pH.
I am not familiar with the procedure used, so I can't give you a lot of ideas.
