I am working with oral cancer cell lines and trying to find the expressional changes of HIF1alpha under different experimental treatment. I am getting intense band of HIF1alpha in the western blot for the whole cell lysate under normal cultured condition( 5% Co2 and 37 degree incubation) without an induction of HIF1 alpha. When I am treating the cells with the chemical inducer of hypoxia (Cocl2 with the dose range of 50-250 micromolar for 24-48 hrs), I am not getting any significant increment of band intensity of HIF1alpha in the western blot. Since we don't have the facility of hypoxic chamber so as per literatures we are using Cocl2 for HIF1alpha induction. Can you please explain why I am already getting enough HIF1alpha expression in normoxic cultured condition and there is no significant induction in the chemically induced hypoxic condition?
