Hello everyone! I am doing half strip test for Lateral low immunoassay.
Here is my detail information about the experiment:
1. Size of Gold nanoparticles that I used is 15 nm.
2. GNPs solution was replace with Borate Buffer 0.01 M pH 9.0
3. Dilute MAb 0.1 mg/mL, incubate for 45 min
4. Add BSA 1%, incubate for 45 min.
5. Resuspend in borate buffer 1 mM pH 9.0
For immobilized antibody:
In the test line, I put my capture MAb 0.1 mg/mL
In the control line, I put my capture PAb 0.1 mg/mL
I have tested with varied MAb 0.1-0.5 mg/mL in the test line but the test line not getting colored even I applied antigen 10.000 ng/mL
For the control line, with PAb 0.1 mg the test line was getting colored.
Any suggestion would be very helpful for my experiment.

For Gold Nanoparticles, i didn't buy from any supliers. I synthesized that GNPs by my self.
