I am working with drug activity on familial ALS model cells. I am using mouse Neural stem cells which express the mutated human SOD1 protein. I did western blot with the protein extracted from the cells. I used SOD1 goat polyclonal antibody raised against a peptide mapping at the C-terminus of SOD1 of human origin. The antibody manufacturer recommends it for the detection of SOD1 of mouse, rat and human origin. But when I did western blot I got a band for the mutated human protein but I didn't get any band for the native mouse protein. Can anybody help me with this?
Blame it to the antibodies (often they are not specific or may losse quality due to the fact, that new charges were not further tested by providers.
