I am trying to set up MTT assay/XTT assay for primary fish cells. But it doesn't work very well. first of all, I ask you share with me your experience about the estimation of toxicity effects on this kind of cells. and also, why MTT assay doesn't work in wave length 570 nm and why in some papers they mentioned that the efficient wave length is 490 nm for this kind of cells?

Absorbances in MTT assay should be read at 550-570 nm (L1) and 620-650 nm (L2). Final optical density is calculated as OD=L1-L2.

My MTT assay work in wave length 570 nm for RAW264.7. I suggest, you try with diffearant wave length. I think, it is about type of cell. but I think. and It is good change for Incubation and rapidity pepat.
