When I started to study the cell culture I have a question, that is why should we add serum media after trypsinization process in cell line established process?

In the trypsinization process extracellular proteins are digested, which leads to the detachment of the cells from the bottom of the culture vessel. Cell culture medium with serum ist added to inactivate trypsin, otherwise the ongoing proteolysis would lead to cell damage. Serum contains many protease inhibitors, which are stopping trypsin, mostly alpha-1-antitrypsin.

The serum has natural protease inhibitors which will neutralize your trypsinsinization. This will keep it from harming your cells if left on for too long.
