I converted differentiated cells into round shaped de-differentiated cells in F12/DMEM medium. As soon as, I transferred them to maintenance medium (TESR-E8 medium) they started changing their shape and have long edges like arm. Any suggestions to keep them in round shape?
I first coated my plate with corning matrigel (1:100 dilution) and then seeded cells into complete TESR-E8 medium. After 18 hrs, they started changing their shape. Any suggestions please!!
I never used TESR-E8 medium, but surely coating with matrigel favor cell adhesion and consequent "arm" formation. definitely you need to change culture condition if you want maintain cells in undifferentiated stage (brw, the shape itself is not a good marker for differentiated/undifferentiated stage). there are commercial media for Cancer stem cells or normal stem cells you can try. coating with anything will induce adhesion and arm formation in most of the case. good luck
You have not provided enough information about either cells type for anyone to properly speculate about the condition of your cells. Is a round shape actually a sign of better or correct cells?
In general though, such "arms" indicate that the cells are happy with the substrate.
