On the way to iPSCs. The targeted cells started to become circular (indication of dedifferentiation). Before I go for their confirmation as iPSCs they started to detach from the plate surface. Now, I am confused that what should I do? Should I subculture them on new (coated) plate? Or should I wait for them to make dense circular network with clear edges by ignoring the detachment aspect? If I need to subculture immediately then what kind of coating I should use at this step when the cells are round but not iPSCs (probably)? Gelatin-coated, Matrigel-coated or inactivated Feeder layer coated? Thank you.
