I am screening a protein-peptides interaction in thermal shift assay. I observe that few peptides are giving rise in ΔTm > 2.0 °C which is considered as potential hit as they seem to stabilize the protein upon protein-peptide interaction whereas few peptides are lowering the ΔTm < 5.0 °C. Can anyone give explanation what could be the issue that I observe decrease in ΔTm or can these peptides be considered a potential hits for further studies.?
