PSD-95 is postsynaptic protein, so why it has major impact on mEPSC frequency for which presynaptic structures are expected to be responsible? Strangely enough, overexpression do not enhance amplitude considering the increased number of AMPA receptor puncta.
Dear Jiahao, possibly i'm wrong, but I think the increase in mEPSC frequency perhaps doesn´t result from a presynaptic modification, but rather it could reflect the conversion of silent synapses to functional synapses. This would be in good agreement with the high proportion of silent synapses found in PSD-95 KO mice (J. C. Béïque et al., PNAS, 103 (51). 19535–19540, 2006). With regard to the amplitude, although mean values are not different, cumulative distribution for neurons overexpressing PSD-95 shows a shift to the right which indicates an increased incidence of high amplitude events. I think a KS test should report such difference.

Thank you for your hint. It is true that PSD-95 has a mixture effect on both transmitter release and postsynaptic structure.
On the other hand, I have no idea how PSD-95 in postsynaptic cell changes presynaptic function, in somehow retrograde manner or homeostatic way. Could you help me on this?
