I am trying to do plaque assay for rotavirus. Though MA104 is widely used for rotavirus assay however vero cell is also recommended. I am trying with vero cell.

But my vero cells detach after few hours incubation with overlay media. 

I maintained some steps not avoiding detaching. 

- Confluent monolayer before infection. 

- Minimum time gap in every steps in infection.

- Cells without overlay media remain okay and show usual rotavirus CPE.

- Carboxy methyl cellulose is used as overlay media. M199 and CMC ratio 1:1. 

- I am attaching control (only cells with overlay media) well image for understanding. You can see vero cell are not in their usual shape.  

Without solving this, I can't develop plaque assay. Requesting your expert advice.
