Hi I have prepared 8% w/v of SF-HFIP solution and started to proceed with electrospinning, I used 18g needle with 1.2ml/hr flow rate and 20KV, but it is getting clogged and the patch is not forming, what might be the problem?
There are a number of factors that can cause this issue. You would have to double-check your operational parameters. If they are the same operational parameters you have been using, then also check the environmental conditions. If the environmental conditions are not constant or are same as what you have been working with, then also double check the properties of the solution
Your hfip solution evaporates fast – before it reaches collector. You can adjust your flow rate, distance to collector/voltage or reduce silk fibroin concentration. Other parameter – room temperature. If it is very hot or the fume hood flow is too high it also can cause fast evaporation
Silk fibroin in HFIP can clog due to rapid solvent evaporation at the needle tip, especially under high voltage. HFIP is volatile, and 20 kV may accelerate drying. Try reducing the voltage slightly, lowering ambient temperature or humidity, or adding a small amount of less volatile co-solvent like formic acid. Also, check if the solution is too viscous or aging (gelation), and clean the needle frequently.
