Why my recombinant cytokine could not rescue the migration ability of cancer cells?

Hello, I measured the concentration of cytokine in supernatant of CAF (SC&KD group), and defined that cytokine concentration was decreased about 5 ng/ml after gene knockdown in CAF. And I applied transwell to perform rescue assay: I set cytokine concentration gradient for 5ng/ml, 25ng/ml and 50ng/ml. But none of them rescued the inhibitory effect of CAF with KD to cancer cell migration. I could not figure out what the problem is. Could you guys help me, please?
