We are trying induced diabetes in Wistar rats using Streptozotocin. But none till now are diabetic.
Protocol used
12h fasting
60mg/kg of Streptozotocin in 0.1 M citrate buffer, pH 4.5 (ip)
prepared just prior the administration and mantained in ice.
are there another tip for this protocol?

I suggest you retry STZ (solved in cold injectable distilled water) injection at the doses of 45-50 mg/kg. after 72 h you can check rats FBS. In my experiences, this protocol lead to diabetes (FBS 450 and above) in Wistar rats.
Fasting continue after STZ injection (8h) , animal weight(180-200 is appropriate) and injection volume (50 unit is excellent) are important.

Prepare STZ in an ice cold citrate buffer. STZ is light and temperature sensitive. So try to prepare in a dark atmosphere if possible.

Streptozotocin-Induced Diabetic Models in rats is after a single i.p. injection of a freshly prepared solution of STZ (65 mg/ kg body wt.) in 0.1 ml M citrate buffer (pH4.3) after 18 hours .so the larger dose of STZ (65mg / kg) will be proper.
Alternatively, try all the above steps in non-fasting animals. In that case no sucrose in water is needed. We have done it many times and there's a 75-80% success in rendering animals diabetic.
I will advise you try to ascertain the source of streptozotocin.
Also make sure the rats are not too young. Young rats are very resistant to streptozotocin. If young, try older rats (3-6 months old).
Also make sure you do not administer streptozotocin more than once to each rat.

2-3 months old is a usual age for this protocol. Resistance to streptozotocin (STZ) is noticed to animals younger than 2 months old (Masiello and Pergamini, 70's). According to our experience as gained in several pilot studies, in case of failure to render animals diabetic, second administration of STZ after 2 weeks, (the way I described it above) is just fine without causing any problems to the animals.
