After preincubation step (pH 10.5) muscle samples melting from the slide.

Hi Mindaugas,

That is probably associated with the prolonged baths and temperature. I suggest you to use some gelatin or polylysine in the glass slide where you are going to put your tissue sections to keep it there during washes and incubation periods.
