Hello, actually I’m working on human mesenchymal stem cells. After a process of expansion, in the 9th passage I found a weird morphology in my cultures, but i don’t know what is happening with my cultures. I am attaching the images of a healthy culture, and the weird ones, some suggestions for me?

Human MSCs subjected to extensive in-vitro passage can undergo morphological, phenotypic, and genetic changes. These properties can also be modulated by the medium composition employed to expand the cell population. Senescence can also affect MSC characteristics and functionality.
In the paper attached below, the investigators observed that MSCs cultured in the appropriate medium maintained their normal spindle shape in the early passages up to P-5, which at later passages exhibited less concordant cell morphologies with some of them possessing irregular flattened geometry and enlarged size.

I agree with the previous answer. I would like to add that normally ageing MSCs change their morphology and become striated (photo). Also, the cells practically stop proliferating. However, the MSC in the photo looks rather strange. Could the culture be contaminated with other cells or mycoplasma?
