I performed laminin staining for ko mice and wt mice 10 days post-injury in the frozen section. the pictures show overlapping ( indicated with a red arrow) both in wt and ko so it isn't individual fiber (shown in the yellow circle). just I wandered if this profile indicated for something ? and also the result of laminin analysis will be affected by those overlapping, won't it ???

Hi Nour,
After injury (like CTX injection, for example) existing damaged fibres are getting removed by macrophages, which go underneath the lamina. Also muscle stem cells (which are located on the fibres under the lamina) proliferate and differentiate into new fibres afterwards. I speculate, that such small fibres inside one big one represent the following situation: "old" damaged fibre still has lamina around it, but muscle stem cells already proliferated and fused together and started forming new muscle fibres (so you see newly formed fibres, surrounded by new lamina).
