Dear all,

I am trying to express my protein of interest (aggregation prone) in a soluble form and decided to give a try to a periplasmic leader sequence. I was hoping for disulfides to form correctly in the bacterial periplasm and so to get native, soluble protein. I am using BL21(DE3) for expression, as most of us.

However, after adding the pelB-leader sequence, the expression is still completely in inclusion bodies...

Should I try other plasmids or other leader-sequences?
