Hello! I've been having trouble with staining lung and pancreatic tissue. While doing H&E, tissue partially (or completely) detaches. Sometimes, the tissue folds on itself as well. Not sure why this is happening. I've been using frosted plus slides and usually bake my slides (60.7C) and/or leave overnight to dry.
Sections are cut 3μm thick with a manual Leica microtome
are they frozen sections or paraffin-embedded. In any case, may be very very gentle in washings since sections are very thin and tissues are very delicate too.
They’re embedded in paraffin. I’ve noticed wrinkles on the tissue as well when cutting. Thank you, I will try that.
Probably microtome shaft has some issue, even your blade may be new. May be try other microtome or get it inspected by a specialist. The detachment on slides have lines that shows that microtome issue
Try reducing the xylene immersion time. try to clear the samples as quickly as possible in xylene.
