ligase extracted from E.coli is used to seal the dissimilar ends of a DNA molecule -DNA ligase. Is ligase is the only enzyme to seal the dissimilar ends of a DNA molecule or any other enzyme we can to seal the dissimilar ends?
There are numerous ligases other than E. coli DNA ligase that are used to join DNA's together. NEB has a good selection chart (linked). I'm not sure if this answers your question, though.
We are all somewhat confused by your question. If your DNA molecule has blunt ends then it should not matter and any ligase that acts on ds DNA should work. If there are single strand overhangs, then it is not likely that you can ligate dissimilar ends since the single strand overhangs will not anneal.
