It is recommended but it is not necessary, especially if you avoid using the well on either end of the gel. As Omar states, it helps avoid lateral diffusion which may create a smile but I hardly ever see this problem.
What happens if the media is double autoclaved immediately after the first autoclaving? I am going to inoculate the Trichoderma; will it affect the sporulation of the Trichoderma?
