Hello everyone. I have a question regarding CFSE proliferation assay with mice. My goal is to see CD3, CD4, and CD8 populations. Why do I keep getting one peak, as if there is no proliferation? I even stimulated the cells using ConA. When I check the the activation marker for the cellular surface, the population is activated. This is my method:
1. 1x10^6 cells in 1 ml per well (Grew in 6 well plate)
2. 5ul of IL-2 daily (60 IU)
3. Staining according to protocol by manufacturer (CellTrace CFSE Invitrogen)
4. 72 hours stimulation
