I am facing problem in making of separating/resolving gel. After polymerization of separating gel when I remove IPA and pour stacking just after couple of minutes these unusual patterns form in separating gel. I really not understand that why they are forming and what are they, bubbles or something else because after overnight storage of gel at 4 degree they disappears by own. And when I run gel they can be visualized. Please if somebody faced same problem kindly help me in sorting out my problem.

please tell me if these patterns cause any hindrances during sample running?

Are they present in separating gel or between the wall of plate and gel ?

Why these patterns are only appearing in separating gel and not in stacking gel?

Are they forming due to pipetting of gel mixture?

*I made 10% separating and 4% stacking here.

*freshly prepared 10% APS and fresh TEMED.

*10ul TEMED was used in separating gel making.
