I am using an extract of pretreated lignocellulose at differing percentages (100, 75, 50, and 25%) as a carbon source replacing glucose in minimal media (normal composition is glucose, 10g/L; magnesium sulfate, 0.2g/L; sodium chloride, 0.5g/L; peptone, 2.5g/L; yeast extract, 2.5g/L) for the growth of Bacillus spp. for PHA production. I added 1.5% inoculum size of Bacillus spp., and after 48 hours of incubation still no growth. What is the possible reason and how can I solve the problem?

P.S.- DNS confirmed the presence of reducing sugars in the extract.
