If you are using a coupled enzyme detection system, there may be a lag phase at the start until the intermediate (the product of the first enzyme/the substrate for 2nd) builds up to a sufficient concentration. This lag phase may be reduced by increasing the concentration of the coupling enzyme relative to the primary enzyme, or changing the conditions to better favor the activity of the coupling enzyme.
