For immunohistochemistry tissue samples are normally mounted on slides coated with poly-l-lysine. But these paraformaldehyde fixed tissues keeps falling of newly coated slides. The thickness of the tissue is 16microns.
Hi Yuganthini, it might be that yout sections are to thick for poly-L-slides. Try superfrost plus slides from thermofisher or any other company who sells labware. Mount your sections on this slides let them air dry overnight and start proceeding your IHC protocol.
I use to mount 16microns thick brain tissue, and it did work. Its just recently that it has started to be an issue. I normally dry my slides at 37C for about 2hrs max. So maybe I should try drying my slides overnight.
Paulina,
Thank you for your reply. It would be great to get the DIY protocol for silane slides.
