I am currently trying to generate MCF-7 stable cell line transfected with LC3B shRNA plasmid (puromycin resistant). After shRNA transfection and antibiotic selection (puromycin selection) (for selection of stably shRNA transfected cells), I have picked different colonies and amplified them in growth media containing puromycin. I have done western blot to measure LC3 protein expression in those colonies.
In all the shRNA transfected clones, LC3II protein was decreased compared to Control (cells without shRNA transfection). However, the level of LC3I protein in all clones remained similar with the Control.
As far as I know, LC3B shRNA should reduce both protein expressions.
I have attached here the image showing C as Control, lane 1, 2, 3 and 4 showing different clones of shRNA transfected cells.
