I splitted my cell 1in 8 from almost 100% confluence and then they reached 90% confluence 24h later. Is that normal for CHO-K1 cells? I used advanced DMEM/F12 with 10% FBS, 1% L-Glu and 1% PS as medium.
If they were very confluent when you split them maybe they were very densely packed. When you replated them they probably stretched out to make contact with each other. Furthermore, if they were confluent they may have had some cells floating alive because they had nowhere to land. They won't grow on top of each other. When you split them you probably had attachment of the floating ones and the ones you detached. I'm not too surprised with your results. A 1:20 is probably a better idea if you don't want to tend to them every day or every other day.
