Dimethyl sulfoxide (DMSO) is commonly used in PCR to relieve secondary structures when amplifying high GC templates. the addition of DMSO can greatly improve yields and specificities of PCR priming reactions . I do not know exactly what is your purpose with DMSO. If it is investigating the Effects of DMSO on PCR Fidelity, so you will find 5% of DMSO is effective.
you can use DMSO to enhance PCR reaction through eliminate secondary structure of DNA and resolve high content of GC in amplicon also you can use other materials like Betaine, DMSO used with 5% of total reaction and you can use 1M of Betaine
