Hello Everyone,
Recently we bought Hap1-WT and Hap1-Knock Out(KO) cell lines. To confirm KO efficiency I did Western blot.The immunoplot result was conformed It’s KO efficiency,but with the ICC result, I couldn’t differentiate the KO difference. What would be the reason? I have used the same antibody (monoclonal) in western and also in ICC. Please see the attachment for reference.

I am not sure of what I see on the images: is this antibody fluorescence, or DAPI counterstaining?
Depending on the answer, you have either no labelling with the antibody, or similar labelling in WT and KO. Let me know, and we'll try to troubleshoot.
Hi again, I have not included the DAPI staining, those images are from green Channel. I have used the same antibody to stain both cell images to differentiate the protein localization and KO confirmation.
