I am trying to measure calcium levels in hippocampus rat slices, specifically in CA1 layer, using membrane-permeable Fura-2. I am using control slices incubated without Fura-2 but with DMSO, the dissolvent in which I resuspend Fura-2. I can observe fluorescence in my slices incubated with Fura-2 but the problem is that I observe fluorescence in the control slices too. I know that autofluorescence could be the reason, but when I add KCl 1M I can observe and strong increase both in slices incubated with Fura-2 and control slices. Do you know what can be happening? Is it possible that DMSO produce fluorescence? Thank you ;)
