Hi,
I coat my plates with collagen and culture my cells (fibroblastic) for 7 days before further analyzing them for immunofluorescence or trypsinizing. I have recently observed that my cells respond to Dulbeco's PBS (no Ca/Mg) by detaching from the plate within 2-5 mins (see attached image). I wash them gently with DPBS so that they probably don't interfere with trypsin/PFA.
Also, while media changes during day(D) 1, 3, 5, I do wash with the cells with the same DPBS and they are ok. The problem is not my media or the DPBS because other cultures with the same cells are fine.
My understanding is that the cells are degrading the coating within the time span of 7 days and so this is happening? If so, does anyone know if increasing the concentration of collagen while coating help the cells to not detach?
