Hi, Prakash, may be TDZ concentration is too high. As sson as you get leaf, you should reduced TDZ in the medium to minimum level. TDZ require only for morphogenesis initiation, but not good for plants growth.
Dr. Pasternak is right. TDZ is effective in morphogenic differentiation but nor preferable further growth and devlopment. Addidionaly, it is highly efective at very low concentrations such as 0.1 and less for many herbs.
Hi Prakash. TDZ in tissue culture experiments has to be used very cautiously since if not used properly, it is a horrible supplement! Subculturing again and again with reduced amounts of TDZ once callus initiatin takes place sometimes helps. Please be aware that sometimes results with TDZ are even misleading!
As we know, TDZ stimulates the production of phenolic compound in the explants and higher concentrations intensify this effect. The response of the explant to the wound is also the production of ethylene and phenol. Therefore, my recommendation is to pretreatment of the explants (before culture) with ascorbic acid or PVP and also to add 0.5% PVP to the culture medium containing TDZ and and finally sub-cultured in a hormone-free medium as soon as possible.
