I accidentally set the initial denaturation at 95 degress for 1 min, but It should have been for 5 mins. I realised it after I started the program while it had reached denaturation step at 95 degress only in cycle no. 1. I stopped the program and corrected the mistake and again restrated it? Will it affect my PCR amplification. My template is highly GC rich
