I'm going to do a dyalisis in vitro release study with 6-8kDa dyalisis bag of PLGA nanoparticles loaded with Quercetin. I want to prepare 5 mL dyalisis bags with a quercetin concentration of 3mg/mL, and I would like to know which approximate volume should I choose to immerse the dyalisis bags in.

The volume of the acceptor compartment should be 6-10 fold the volume of the dialysis bag (30-50ml in your case) to provide the driving force your drug will need to cross the membrane. The molecular weight cut off of the dialysis membrane is also a key parameter in the dialysis method, maybe you could choose an higher MWCO for your membrane, mimimum 12-14KDa

It would be related to the levels of sensitivity in the detection method you using and how frequently you taking subsamples and what volume of subsamples and over what time. Generally you would do a much larger volume than what in your dialysis cassette. So likely you want a x 20 or x50 or x 100 volume of buffer.
