Since suspension cells have to be spun down before removing the media, I am just wondering weather there could be any advantage of using round bottom 96 well plates rather than flat-bottom 96 well plates?
Round bottom plates favor cellular contact. It then better for culture involving this type of cell to cell contact. It is also better for cell culture involving < 50,000 cells. Over 100,000 however, flat bottom are better as in a round bottom plate the cells will aggregate and form layers (like a pellet). Flat bottom are also of course recommended for adherent cells culture. If you must use flat bottom, you can still harvest your media, transfer it in a round or V bottom, spin and get rid of residual cells.
