i have started my experiment in greenhouse, which is the formation of adventitious roots applying different hormones and different concentrations, through cuttings propagations. So can anyone suggest me that, which time period is suitable to take samples of adventitious roots for the extraction of secondary metabolite

At maturity in , in vivo experiments ? I suggest you to collect the literature pertaining to your experimental plant and see the harvest time for simplification of process?

To collect the roots of any plant will damage the plant, so it should be ascertained that fruits and seeds are mature before harvesting the roots of plant. Other point may be what time of the day is suitable, as per me it would be harvested early in the morning if you have to collect the secondary metabolites etc. Traditional people also prefer early morning for extracting the plant parts, as at that time there is no sunlight, which can have any impact on it.
Satvant kaur Saini thanks for this precious compliments, but i am not collecting roots from the mature plants, for adventitious roots i cultivated the cuttings in the greenhouse.
OK for most of the plants best time of collection already studied and you can get it from available secondary literature. Otherwise, in green -house or laboratory conditions one has to analyze the secondary metabolite content at specific time interval and find the suitable time period for that specific species. As these parameters are site and species specific.

Yes, you have to design a plan for 2-3 time periods and with minimum number of samples to be collected at each time. Analysis of this study will give you the right time of extraction to get maximum metabolites.

It rather depends on what you want to measure; root growth? Uptake concentrations? Speed of breakdown? I would suggest you either use 7 or 14 days OR as Anoop Kumar Srivastava suggested when the roots stop increasing in weight.
Jonathon Harrington i am going to check the total flavonoids and total phenolics compounds and also checking the effect of hormones on metabolites. the samples is adventitious roots,.
