Sir what is the procedure for sample preparation for AFM?

sorry, I just have done the SEM test, but seen others detect the biofilm with AFM.

For SEM, the samples were fixed, Gradient elution using ethanol,and drying.

using SEM how can i measure the thickness of biofilm?
