I have chemically synthesised spherical shaped iron oxide nanoparticle bearing a size of 20-25 nm with zeta potential value of -34.6mV in aqueous solution. While performing antimicrobial assay, we have dispersed IONPs in water and further sonicated it for 15 mins. The particles then gets precipitated in the agar wells, hence no proper diffusion zone occurs.

What can be the proper methodology to achieve a good dispersed IONPs so that we can used it in a model study?

To be in note: Coating of IONPs not appreciated. The solvent to be used should not affect the immune system of the model study.
