Dear collagues. We planned a new study to evaluate effects of resveratrol ana CAPE on pregnant rats in endocrine disorder. However, because of low water solubility we cannot transffered all the LD50 dosage. could you answer please which solution is more preferrable for solving Resveratrol and also CAPE?
Why would you be dosing them at LD50 anyway? It's a splendid way to induce liver CP450 detox routes that aren't biochemically relevant (and may be misleading, see controversy over safrole for example). But if you must, give by gavage in oil suspension (but see http://www.procedureswithcare.org.uk/oral-gavage-in-the-rat/ ), since most organic solvents for polyphenolics are toxic. You could also try topical DMSO solution -- e.g. under an occlusive patch on bare skin -- but the DMSO will significantly dampen your inflammatory pathways on its own, and your dosage would be uncertain (without confirmatory blood tests).
However, Cansel, this question does not appear to be for an in vitro cell-culture model, but rather an in vivo study. Also, while ultrasonic treatment may increase the rate of dissolution, it won't increase the solubility as such. Perhaps beta or gamma cyclodextrin could be an oral gavage carrier, but that wouldn't necessarily increase the systemic availability -- that you might check with radiolabel-spiked study compounds, enabling lower level blood detection (HPLC + radiodetection).

Dear Stanton De Riel thanks for your interests to my question. As you think that this study both in vivo and even fetus toxicity study. But yours advice beta and gammadextrin may effectiive. I will try at least one group to get information about effects of this chemicals and share my data in this platform. Best regards
By the way, Murat, polysorbate-80 (polyethoxylated sorbitan monooleate) and "Cremophor" (polyetholylated castor oil) are also used to solubilize fairly-large-molecule antineoplastic drugs, which may then be diluted and injected IV.
