HER2 and HER4 are considered to be related to xCT (cystine amino acid transporter essential for the synthesis of GSH) at the cellular membrane. Why not perform coimmunoprecipitate experiments to show the interaction between HER2 and xCT or HER4 and xCT?

Several publications described RTK versus ROS relationships, including members of ErbB family. In fact, the generation of H2O2 itself might be involved in the activation of RTK in cells treated by chemical compounds. There is a confusion with your reference. Do not mix the gene ROS1 (ROS proto-oncogene 1, receptor tyrosine kinase) with ROS (Reactive Oxygen Species). No relation has been shown between ROS1 and ROS.

Dear Vehary! Thank you for a delicate reminder of the conventional view. You're right. However, we recently check the hypothesis of the involvement of free radicals in the processes that are associated with a change in the functional state of tyrosine kinase receptors. It seemed to me that this idea is quite useful to a young scientist. The basis for our hypothesis were the articles on this subject (Please see attached). Vladimir

It was a good ocassion to talk on  RTK & ROS interactions, that is challenge in biomedical research and drug development. Just one more point that should be taken into account. While cells are treated with pharmacophores, their action might be also related with a formation of concomitant electrophilic intermediates, and which independently on ROS also attack proteins (see Discussion in our paper, above). However, EGF binding to EGRR differently generates ROS, through Nox2, as shown in an excellent paper of Carol's group: http://www.nature.com/nchembio/journal/v8/n1/full/nchembio.736.html
