At the risk of some false positives, I want to maximize initial detection of a subgroup 1a rhabdovirus. Which has the higher sensitivity, semi-nested RT-PCR with degenerate primers or rRT-PCR?
though both are good choices, based on my experience, i always feel real time RT PCR has better sensitivity than conventional RT PCR , also has better specificity due to less handling .
on other hand Semi nested RT PCR has better sensitivity than just first round RT PCR. But then as handling increases during reaction set ups, chances of contamination also increases.
