Temperature, moisture and oxygen, but light is usually not necessary
Seed soaking time and sterilization methods may vary for different species. It depends on size, age and permeability of seed coat. For sterilization UV radiation can be used or you may follow another methods (for e.g. temperature treatment, etc.). The parameters used for seed germination experiments may be days of initiation of root development, days taken for germination of seed, initiation of first leaf, germination percentages, size of root at a particular day, etc. These parameters can also be useful for comparing seed germination experiments for various different species. One can also study effect of different physical (temperature, light, etc.) and chemical factors on seed germination of different species.
for example you can also considered days of root development, Germination%, Germination speed, survival rate, shoot height, diameter at root collar, root length, shoot/root ratio, number of leaves.
Dear Bilal, various parameters are available for seed germination experiments out of them you can choose the parameters which determine the completeness of germination and speed. ie. Germ%, R50, FDG, LDG, MGT, Timson Index etc. For details refer the attached paper
There are different methods of seed priming, you can follow one of them for improved germination. You can compare field germination and Controlled germination.
there are many methods to test the seed germination and viability. Which method should be adopted is depends on the size of the seed. the best parameters you can assess the seed germination capacity are seed size, seedling dry weight, speed of germination, Seedling growth rate (SGR), Seedling vigour index (SVI), electrical conductivity of seed leachate most important to compare the seed germination capacity is field emergence percentage

It is always good to know a little about the biology of the plant. Many seeds will have dormancy and it is important to break this before looking at germination rates etc. The best way to break dormancy will depend a lot on the seed type etc. For example, if it has a hard seed coat, you may want to look at scarification through physical or chemical means. It is is a temperate species, cold stratification would be pretty standard. Understanding the plants life cycle will help a lot with this.
However, not all seeds are dormant, so you could initially try germinating them. If the germ rate is low, test the viability to determine if the seeds are dormant or dead.
Once you ensure dormancy is not an issue, I would proceed as suggested.
