I am trying to sequence a 10kb DNA amplicon. The amplicon was generated from cDNA with long-range PCR.

I want to sequence the 10kb DNA using Oxford nanopore but I am not sure which protocol should I use and which sequencing kit is best for me!

I have the Ligation Sequencing Kit SQK-LSK110. There are many different protocols in the ONT website related to this sequencing kit such as:

  • Genomic DNA by Ligation
  • Lambda Control Experiment
  • Amplicons by Ligation
  • Low input genomic DNA with PCR
  • PCR barcoding genomic DNA
  • PCR barcoding amplicons
  • PCR barcoding (96) genomic DNA
  • PCR barcoding (96) amplicons
  • Premium whole genome amplification
  • Exome sequencing
  • Sequence capture

Which one should I use? is it the "Amplicons by Ligation" protocol?

According to the nanoporetech website (https://store.nanoporetech.com/ligation-sequencing-kit110.html) this sequencing kit should be used for singleplex samples. Does this mean that I can not sequence multiple samples at the same time using this kit?
