I have a aqueous solution of an organic compound, initial 400 ppm. later on after treating the solution the organic compound is degrading. let us say, I am finding 200 ppm, 100 ppm, 50 ppm and 0 ppm after a time of 1h, 2h, 3h and 4h respectively. I am giving this sample as treatment into E.Coli bacteria (LB media etc.), and finding more colonies in 0 ppm treatment and no colony in 400 ppm treatment. What does it mean? which sample is more toxic to E. Coli.? Comment if information is unclear. Thanks

I want to do toxicity test for different concentration of phenol aqueous solution e.g. 400ppm, 200ppm, 100ppm, 50ppm, and 0 ppm phenol. For that I am taking 5 petridishes having LV Agar media and introducing 1 micro liter of E Coli bacteria solution along with 1 ml of above phenol solution. After 24 h of incubation at 37 oC I am finding more colonies in 0 ppm petridishe and no colony in 400 ppm petridishe. What does it mean? which sample is more toxic to E. Coli.?

Bacterial colony will be formed only by the living cells, the higher the colony amount, higher is the number of living cells.
In your experiment, you got more colonies in 0 ppm, that means there is no toxic material is there and bacteria can grow easily, whereas, in case of higher phenol concentrations, bacteria are being killed and no colony was observed.
From your data, it is simple that 400 ppm is more toxic than 0 ppm. This is very normal.
