Dear All, mirna primer showing some problem in the melting curve? any idea why? As attached is the melting curve. The forward sequence is obtained from miRBase and reverse primer is universal.

Hello Gil,

Will it be possible to use spoken miRNAs from others species than Gallus Gallus (Ath, cel?). Those will also help you to validate the different steps of your experiments (RNA purification, RT and qPCR)

If not, you may find the most invariable miRNAs in your data set, independently of your experimental conditions and use them as Reference .
