When studying reporter gene expression in mice by isolating cells that express a reporter gene (let’s say GFP, dsRed or other fluorescent reporters) and subsequent FACS analysis/sorting, would there be any specifically well-suited organ with regard to the ease of primary cell isolation, FACSability (autofluorescence,…) etc.?
In other words, if the biological context and genetic manipulability would not be important at all, which would be the organ/cell type you would prefer in terms of the ease of cell isolation and FACS? (Blood-derived cells are not an option). Thanks for sharing your experience!
I believe spleen lymphocytes (T or B) are the easiest.
(i) The organ is very easy to isolate and to process in order to get cell suspension;
(ii) These cells are the major cell type population in this organ;
(iii) Lymphocyte markers are very well characterized so you can easy use (CD3, CD19 antibodies).
Spleen and BM are indeed easiest as the cells are nice and small and only mechanical dissociation is needed. After that I guess liver, pancreas and ovaria/testes, then kidney, then gut and lungs. Skeletal muscle and heart are difficult and I don't know about the brain, I don't think it's possible.
