Methods:
1. s.c injection: 0.5ml/kg bw, thrice/week upto 4 weeks
2. i.p injection: 0.5ml/kg bw, twice/week upto 4 weeks
3. oral gavage: 0.5ml/kg bw, thrice/week, upto 4weeks
I have tried the 1st method but got poor result regarding fibrosis development.
I have read several papers on the effectiveness of the intraperitoneal method. This is the method I will be attempting soon, with the approval of my institution's IRB.

We tried fibrosis induction with CCL4 in Balb/c as well. Balb/c mice are very susceptible to CCl4 with quite high mortality rate. With 0,2 ml/kg Body weight we reached after 6 weeks a strong fibrosis. Good luck!
i tried CCl4 and thiocetamide but their action is through direct cytotoxic effect followed by recruitment of inflammatory cells followed by fibrosis but i think these models are no suitable for studying cytokine profile but i also tried a model of fibrosis utilizing Concanavalin A as a model resembling viral hepatitis as it recruit inflammatory cells first that produce necrosis and degeneration so the model type depends on the aim of the research

I agree with A. El-Karef that it depends on what type of fibrotic mechanism you are studying. For instance, use of high-fat and high-sugar intake can lead to fibrosis after a stint of steatohepatitis. This would be a more useful model for research into NAFLD/NASH pathology. CCl4 is more useful for alcohol-induced and some hepatic neoplasm pathologies.

The answer is depending on what you need really. Septal fibrosis requires over 6-8 weeks by dose dependent manner. IP develops it faster but often has higher mortality and scar formation on liver surface which is not relevant. I used to feed 3 times/week for 8 weeks to establish severe than septal fibrosis. Balbc strain could be more succeptible for liver disease than B6 strain. Please keep them in mind that
- to harvest at least 36 hours after the last feeding to avoid having acute hepatitis
- to use over 25g mouse
- to harvest not later than 3 days after the last feeding, of course, it reverses.

we have been planning to carry out the chronic model system to test our drug compound. Now we've decided to go for the i.p. for 8 weeks: 0.5ml/kg bw. again in Balb/c and will harvest after 24 hr starvation. after 6weeks we will assess one from each the group if there is any systematic development of fibrosis.
